Structure-fonction des complexes nucléoprotéiques des ADN topoisomérases
RESPONSABLE DE SOUS-GROUPE
Nous nous intéressons à la régulation de la topologie de l'ADN dans les bactéries et chez l'homme, en particulier la famille des ADN topoisomérases qui forment des complexes nucléoprotéiques et catalysent le relâchement des surenroulements de la molécule d'ADN dans la cellule.
Nos axes de recherche portent sur les études fonctionnelles et structurales des complexes cellulaires des ADN topoisomérases ciblés par des composés thérapeutiques utilisés dans les traitements anti-cancéreux et anti-bactériens. L'objectif principal de ces recherches est d'identifier et d'étudier la relation structure/fonction des complexes associés aux topoisomérases et ciblés par des médicaments grâce à une combinaison d'approches fonctionnelles et structurales.
Projets en cours
- Structure/fonction des complexes chromatiniens des ADN topoisomérases humaines (TopoII).
- Etude de l'impact fonctionnel et structural des modifications post-traductionnelles des isoformes des TopoII.
- Structure/fonction des complexes nucléoprotéiques de l'ADN gyrase bactérienne. Voir notre dernier article publié dans Science en 2024: ici
- Etude de l'impact de la topologie sur la transcription (en collaboration avec l'équipe d'Albert Weixlbaumer-IGBMC)
